Tissue and Cellular Therapies


University of Iowa Hospitals and Clinics DeGowin Blood Center’s Tissue and Cellular Therapies is comprised of two distinct sections.  The Hematopoietic Progenitor Cell Laboratory provides processing and cryopreservation of peripheral blood and bone marrow for patients receiving either autologous or allogeneic hematopoietic stem cell transplantation.The Tissue Bank provides human tissue required for some surgical procedures and provides traceability of the tissue from source to implantation.

The DeGowin Blood Center Tissue and Cellular Therapies has received accreditation from the Foundation for Accreditation of Cellular Therapy (FACT) and is registered with the Food and Drug Administration (FDA). It is also accredited with the National Marrow Donor Program (NMDP).  The Tissue Bank has been accredited by AABB and CAP.

Hematopoietic Progenitor Cell Laboratory (HPC Lab)

The HPC Lab provides service primarily for the Adult and Pediatric Blood and Marrow Transplant (BMT) programs in which approximately 100 – 120 BMT’s are performed a year.  The HPC Lab also prepares HPC products collected at UI Hospitals and Clinics, as a National Marrow Donor Program (NMDP) collection center, for transport to other institutions.  The HPC Lab participates in both institutional and multi-center study protocols of both hematopoietic and non-hematopoietic nature.  Processing performed in this laboratory is done under cGMP and cGTP guidelines as the goal is to provide a high quality, viable, and sterile product to help ensure timely engraftment or treatment following a BMT.  This is done through graft manipulation and/or careful preservation of the product(s).

The HPC Lab is about 1,200 sq. ft. with a separate cryopreservation room of about 235 sq. ft. and another 245 sq. ft. of office space.  The lab has two bio-safety hoods, an Isolex 300i immunomagnetic cell separator, a Cobe 2991 cell washer, several centrifuges, a -80C and -20C freezer, a refrigerator, 6 LN2 storage units, a controlled-rate freezer, and five personal computers for data management.

Procedures performed in the HPC Lab include:

  • Product evaluation (cell counts, cell viability, volume, sterility assessment, flow cytometry)
  • T-cell depletion of allografts
  • Tumor purging of autografts utilizing the Isolex 300i
  • Volume reduction
  • Donor lymphocyte infusion (DLI) preparations
  • Cryopreservation
  • Thawing for infusion which may or may not include washing
  • Product preparation for transporting to other institutions

Tissue Bank

The Tissue Bank maintains an inventory of a variety of human tissue required for surgical procedures such as treating burn patients, bone and joint reconstruction, cornea replacements, and cardiac procedures, performed at the UI Hospitals and Clinics.  These tissues need to be stored and maintained at specific temperature ranges.  Bone putty/paste, bone wedges, sclera, facia are kept at room temperature.  Cornea and manufactured skin are kept refrigerated.  Cadaveric skin, whole bone and ligaments are stored frozen (-80C).  Human heart valves are a tissue that requires even colder storage at < -100C.  Tissue is routinely delivered to the operating rooms while maintaining the storage temperature.

The Tissue Bank is about 450 sq. ft. and consists of a refrigerator, 3 -40 to -80C freezers, and 3 cabinets for room temperature storage.  Two of the 6 LN2 freezers in the HPC Lab cryopreservation room are used to store all non-hematopoietic tissue.  The tissue records are kept in a database that allows tracking of each tissue from the time of receipt to the time of implant or disposal.
